چکیده

Mutations affect the genes that encode certain cytochromes P450 (CYP450) isoenzymes are responsible for metabolism of drugs. The aim this study was to determine frequency CYP2B6 (516G>T) SNP in Burkinabè population. Genomic DNA extraction from blood performed by GenJET® according manufacturer’s instructions. Genotyping cytochrome variant alleles using predesigned primers. amplification carried out PCR while differentiation between after digestion with restriction enzymes electrophoresis. c.516G>T successfully analyzed 92,38% (291/315) participants. Among investigated individuals, distribution major allele CYP2B6*G 56% and minor CYP2B6*T 44%. results obtained showed a prevalence G 44% T allele. This distributed as 31% GG genotype (reduced dosage required), 51% GT (normal range), 18% TT (high drug toxicity). presence rapid poor metabolizer phenotypes studied sample shows need additional studies better assess their impact on Burkinabe people.

The naturally occurring cytochrome P450 (P450) 2B6 K262R mutant of P450 2B6 exhibits alterations in substrate metabolism and inactivation.

The polymorphic human cytochrome P450 (P450) 2B6 is primarily responsible for the metabolism of several clinically relevant drugs including bupropion, cyclophosphamide, propofol, and efavirenz. Although a number of single nucleotide polymorphisms have been found in the P450 2B6 gene, the influence of these variants on the metabolism of substrates and on the response to known inactivators of P45...

Mechanism of inactivation of human cytochrome P450 2B6 by phencyclidine.

The mechanism behind the observed inactivation of human P450 2B6 by phencyclidine (PCP) has been evaluated over the past 2 decades. The scope of the current investigation was to contribute to the fundamental knowledge of PCP oxidation and perhaps the mechanism behind P450 inactivation. To study the chemistry of PCP oxidation, we subjected PCP to the Fenton reagent. Under Fenton chemistry condit...

Functional characterization of cytochrome P450 2B6 allelic variants.

Cytochrome P450 (P450) 2B6 is a hepatic enzyme of potential importance for the metabolism of clinically used drugs and environmental or abused toxicants. Genetic polymorphisms of CYP2B6 (CYP2B6*2, CYP2B6*3, CYP2B6*4, CYP2B6*5, CYP2B6*6 and CYP2B6*7; wild-type, CYP2B6*1) were found previously in white and Japanese populations. In the present study, the goal was to investigate the effects of amin...

Allele and genotype frequencies of cytochrome P450 2B6 gene in a Mongolian population.

CYP2B6 plays an important role in metabolizing various drugs in common clinical use. Increasing interest in CYP2B6 genetic polymorphism was stimulated by revelations of a specific CYP2B6 genotype significantly affecting the metabolism of efavirenz, an anti-HIV type-1 agent. The present study determined the CYP2B6 haplotype in 100 healthy unrelated Mongolian volunteers by analyzing the genotypes...

Corrections of Frequencies of Cytochrome P450 2B6 and 2C8 Allelic Variants in the Mozambican Population.

BACKGROUND The cytochrome P450 enzymes (CYP) play an important role in the metabolism of many therapeutic agents. The activities of different enzymes exhibit variability in different populations, which causes variations in drug response or toxicity. The CYP2B6 and CYP2C8 enzymes are encoded by polymorphic genes characterised by different single nucleotide polymorphisms (SNPs). Several of these ...
